That's interesting. I read it quite differently.
First woman throws in the unscripted "or uncircumcised" line to go with the list of reasons about the problems of finding a man, and quite obviously throws off her colleague, who goes "Ewww" in response.
The both begin laughing and my take is, in an effort to explain away her spontaneous slip, the first actress laughingly says "What does THAT mean?" - as though she's just downplaying the whole thing now that she's brought her co actress and the crew to laughter.
